Production of Cross-Linked Lipase Crystals at a Preparative Scale

The autoimmobilization of enzymes via cross-linked enzyme crystals (CLECs) has regained interest in recent years, boosted by the extensive knowledge gained protein crystallization, decrease cost and laboriousness process, development potential applications. In this work, we present crystallization preparative-scale production reinforced lipase (RCLLCs) using a commercial detergent additive as raw material. Bulk was carried out 500 mL agarose media batch technique. Agarose facilitates homogeneous crystals, their cross-linking treatment, extraction. RCLLCs were active an aqueous solution hexane, shown hydrolysis p-nitrophenol butyrate α-methylbenzyl acetate, respectively. presented both high thermal robust operational stability, allowing preparation packed-bed chromatographic column to work continuous flow.
